In order to determine if FruR directly binds to the ptsHI-crr operon, a 484 bp DNA fragment encompassing the regulatory region of this operon was used in a DNA band migration retardation assay. This fragment was retarded when FruR was present. The addition of fructose-1-phosphate to the binding reaction prevented the binding of FruR to the pts regulatory region.
