As the concentration of MetR was increased from 0.29 to 36.6 nM, a DNA fragment carrying known MetR operator sites ( sites 1 and 2) was shifted to two bands of lower mobility. At higher concentrations of MetR (greater than 36.6 nM), a third band was observed, suggesting that a third binding site with a low affinity for MetR is also present. Homocysteine resulted in a significant increase in MetR binding to the glyA control region.
